Ultra-deep and Quantitative Phosphoproteomics

Precision Proteomics Value-Add

  • Discovery Phosphoproteomics: Systematic identification of Tyr/Ser/Thr phosphorylation on cell lines, animal models or tissue samples for the identification of kinase substrates, kinase inhibitor targets, and regulatory networks associated with cellular perturbations or drug treatment.
  • Quantitative Phosphoproteomics: Label-free quantification or isobaric labelling followed by LC-MS/MS to determine dynamic and systematic changes in the kinome and phosphoproteome associated with cellular perturbation or drug treatment.
  • Mechanistic Studies including MOA: Systematic and quantitative characterization of signaling pathways and protein phosphorylation perturbed by gene manipulation or pharmacological treatment in cells and animal or tissue samples.

Case 1

  • SH2S –MS/MS identified the largest tyrosine phosphoproteome (as of 2016; 10,030 unique pTyr sites, 36% of which were novel)
  • SH2S –MS/MS identified the phosphoproteins associated with response to kinase targeted therapy.

Case 2

  • SH2S enable systematic mapping of activity of all known Tyrosine Kinases
  • SH2S –MS/MS identified the phosphoproteins associated with COVID-19.
